In 3-4 years it will be the new Vikings Stadium but until then I would say the "must see" spots are divided.
In Downtown check out the Holidazzle Parade in the winter and, in any season, the Guthrie Theater, Mill City Museum, and Main Street/Nicollet Island. Access the skyways to shop, sleep (not kidding, tons of hotels), and eat.
Near Downtown, you must see the Sculpture Garden, Walker Art Center and Loring Park. Further up Hennepin or Lyndale Avenues, you will reach Uptown or LynLake. Close to these neighborhoods are the beautiful city lakes, as well as the Rose and Japanese Gardens.
In South Central, check out the Midtown Global Market or Mercado Central for unique shopping and ethnic eats.
Northeast Minneapolis has an "up and coming" arts district as well as an endless number of diverse and traditional restaurants.
Don't miss our parks. They are all great but Minnehaha Falls offers beauty and history. It is close to the River Road, Fort Snelling and the Greenway Bike Trail.
Theodore Wirth Park offers downhill and cross country skiing and, after the thaw, the Eloise Butler Flower Garden is always in full bloom.

Wow! Thank you for all the wonderful ideas! I didn't know the Vikings were getting a new stadium, so that will be cool. I've heard a lot about the Mill City Museum but still haven't made it there. I'll make sure to put it on the itinerary for my next visit!
